These PLGA nanospheres are extremely densely packed, and do not dissolve well in solution. I have already tried DMSO and release solutions such as SDS and 0.05M NaOH. The protein encapsulated in the particles is mouse IL-12 (p70). We want to know how much IL-12 is inside the particles, but we cannot figure out how to dissolve the capsules.

Perhaps you can consider using dimethyl sulfoxide plus ten milliliters of 0.05 M NaOH containing 0.5% (w/v) sodium dodecyl sulfate.
Hi there -- thanks very much for your answer. I really appreciate it. I have not read the paper from 2016 and will definitely look into it. So far, I have tried the solution you mentioned (as well as the same solution with twice the base!). The nano particles are so tightly packed that they do not dissolve all the way and form clumps.
