Thanks for your attention Mr. Joshi. I want to determine an analyte by HPLC but it doesn't show a UV active property. I can use only UV detector with HPLC device. Therefore I think that I have to use ındirect UV technique. I'll add a UV active reagent to mobile phase to achieve this technique. Which reagents can be appropriate for this purpose for both reverse phase and normal phase HPLC system?
Its important, what you want to analyse. The are different reagent for use as derivatization agent. You can first derivate your sample and then analyse with UV.
I think F. Akbas is right. if any compound can not be detected by UV, then we should go for derivatization. this is usually done before injection. Derivatization reagents differ with the analyte molecules.
Adding an other detector (RI; fluorecence; ELSD; electrochemical;...) to your HPLC system is straight forward. There are USB AD convertors to get the signal in Your PC. This approach will give less problems then derivatization or indirect detection.
I will agree with Fatma and Totan. First try with derivatization,it helps you alot. Just check out if the attachment will serve your purpose. Good luck.
