I am planning to add some comments about how far cancer biology or cancer systems biology could go (key advances) in terms of (1) mechanisms (2) diagnostics ad (3) treatment. Welcome to comment. The ones who have valuable comments will be acknowledged in the review paper.
The mechanisms of programmed cell death induced by anticancer agent should be elucidated. It should be taken into consideration that chemotherapeutic drugs can trigger not only apoptosis but also autophagic or necrotic cell death, or senescence of cancer cells.

Really your facing towards appropriate direction, since we have lot many causes for cancer it is very difficult to understand the actual cause by traditional practice we have. In that concern systems biology can surely help you to identify the cause by analyzing association between the pathways which are playing crucial role in cancer. Pathway analysis is a major approach in systems biology so I hope you can work with pathways related to cancer.
Cancer is actually a systems biology disease as reviewed by Hornberg et al. Here you can stress on the predictive models that can be built based on integration of vast amount of data coming out of -omics. These predictive models will help us make the diagnostic and treatment decisions more informed. The future of systems biology lies in ability to integrate the data from different sources and built a robust predictive model.
