Cleanest results come from in situ or isolated pancreatic perfusion when the organ is infused with medium containing 15.6mM glucose for a fixed period of time (sustained challenge).
@Antonio For an OGTT, thinking of measuring 0 min, 2.5 min, 5 min, 7.5 min and 10 min insulin and glucose levels. WIll that be fine or any other suggestions, please?
