I have been trying to optimize the culture conditions for siRNA-injected prophase I mouse oocytes (CF-1) for 48 hrs. I have been culturing them in Bicarb with 250 µM dbcAMP and I change with fresh Bicarb with dbcAMP after 24 hrs of incubation. However, after 48 hrs the oocytes become very granulated and unhealthy. I would greatly appreciate any guidance or insights on how to ensure the well-being of my oocytes throughout the full 48 hours following injection.
