In general first you need to have pure cultures of the isolates. second, probe differen tculture media to see in which of them the isolate produce itssexual/asexual reproductive structures. Then compare its structures with those registered in fungi compemdium. Thus you know the class and genus to which belong the isolate. Later based on morphometric features of the reproductive structures you can identify thespecies.
First of all, you should use morphological identification methods. Basically, use some media inoculate petri dishes for a week. Then put a fungal piece on the slides examine under a microscope then use with identification key books. If you can identificate for example genus level you should use PCR methods with species specific primers or send any company for sequences.
