We intend to use Herceptin in our lab for in -vitro assays.
We purchased one vial that contains 440mg which is a lot for our needs (and very expensive as well (!)).
Is it possible to reconstitute all of the powder and keep aliquots at -20°C? or at 2-8°C?
Based on the product leaflets, A vial of Herceptin® reconstituted with Water for Injection containing 1.1% benzyl alcohol, as supplied, is stable for 28 days when stored refrigerated at 2°C–8°C. However we would like to use it over a longer period of time.
Can anyone give some idea about the stability and storage conditions of this drug?
Herceptin that you buy from local pharmacy or given for R&D purpose from Genentech is most stable when stored in lyophilized form. According to your need, take the required amount and dissolve (very very gently as these forms dimers and trimmers pretty efficiently) in PBS (pH 6.0 is suitable). Herceptin is highly unstable when stored in any kind of buffers even with supplements (for e.g. histidine) that comes added with Herceptin when you buy them. Hope this helps.
herceptin is antibody, keep powder at -20oC for storage. if you dissolved in solution, pls give additives of 5% albumin, 20% glycerol, at -20oC for storage.
We used to reconstitute according to instructions and then sub aliquot into single use vials (maybe a few uses). Depending on what you plan to do will affect the size of the aliquots. Once we thawed we would store at 4 degrees C for up to 2 weeks. It was usually used up by then. Our goal was to avoid freeze thaws as these may introduce complexes. We stored the remaining aliquots at -80 degrees C until needed. The Herceptin seemed to work find in our in vitro assays under these conditions. The stability listed by Genetech is for human injection. This is quite a bit more stringent than what an in vitro assay will tolerate, but I would still try to avoid freeze thaws and long storage at 4 degrees.
Sorry, I have similar question but it is related to storing herceptin vial at elevated temperatures. We purchase herceptin abroad and it was transported improperly in without thermal protection for about 3-4 h flight (in baggage compartment +5 +15 C ). I wonder if herceptin can be still used for treatment?
