We are testing the anti PD-1(Ketruda) activity on human PBMCS in vitro.
Is there any recommended concentrations that are being used?
I would be grateful for some references in that respect.

In the literature, a often used concentration is 10 µg/ml.
We've tested nivolumab and found activity at all concentrations tested. We did half log steps between 0.1 ug/mL and 10 ug/mL.
I would recommend that you empirically determine the optimum concentration for your particular assay by performing a dose-titration curve. You should do this a a couple of your favorite donors so you know the variability.
Good point Sandra. Different antibodies, different donors, culture medium, always good to test a range of concentrations in your own lab.
