I am working on a project trying to reduce the damage caused by Seahorse XF for retinal / brain organoids for long duration metabolic measurements. The challenges that I face are long plating duration, dissociation from the well plate, averaged measurements across all cell types. Could anyone recommend alternative protocols / equipment?

Your questions sound interesting. What metabolic measurements are you currently using? Do you focus on pH or oxygen measurement in the Seahorse? How long would the measurements cover?
We are developing 3D cell culture systems based on microcavities at the Karlsruhe Institute of Technology. Organoids, for example, can also be generated in these and cultivated over longer periods of time. We are now able to measure oxygen in these microcavities in 3D, i.e. directly on the organoids. Ideas for pH measurement are also in the pipeline.
We founded a spin-off this year (CAVIGEN: www.cavigen.eu), which provides tools for these applications.
