Phosphorylation is the most common post translational modification to control the activity ofProteins. The OH group of serine therionine and tyrosine is the target site. If one wants to study the effect of phosphorylation on function of the target protein, two stratgies can be used: simply mutat this with another amino acid and look for phenotype that is by loss of phosphorylation site. Phosphominetics on the other hands are created by replacing the phosphorylated a.acids by amino acids which appears similar to phosphorylated a.acids such as replacing phospho serine by aspartic acid. This leads to same ohenotype as by the phosphorylated aa.acid in the protein.
