Hi Everyone, hope there is someone on the planet that could help met with advise on the villencre PCR. I do not get it to work. I am currently running a multiplex PCR with MYD88 as an control and everything seem well. However, the cre band is not visible... no amplification... I am doing a touchdown PCR and I also have tried a step PCR but nada... nothing... not even a smear or some sort of none specific PCR product pretty markers and perfect MYD88 amplicons....

CHEAT & Use a KIT !!! Qiagen do a kit Rapid and effective genotyping of Cre transgenic mice. Primers specific for the Cre transgene
sense: 5’- GAACC TGATG GACAT GTTCA GG -3’;
anti-sense: 5’- AGTGC GTTCG AACGC TAGAG CCTGT -3’
amplify a 320 bp fragment.
