Verifying certain UTR's for endogenous microRNA regulation has anyone seen a dependence on the amount of ng transfected for a luciferase-UTR reporter?

Dual luciferase Assay, if saturating cell with reporter, no microRNA regulation observed, more binding site number for a microRNA in a given UTR seems to require more reporter, but less binding site number for a microRNA in a given UTR seems to require less reporter to see an effect of rescue, be it a sponge or antagomir.
