Hi, for detection cellulose biosynthesis in E.coli nissle 1917 I used congored plate, but which condition( temperature and incubation time)is appropriate for just cellulose biosynthesis detection, not for Curli fimbria? for this strain 48hours at 28°C is good or 24 hours at 37°C , or ANY other condition?
What version of Congo red agar are you using? I am currently looking at cellulose production in Cronobacter and have found some recipes for Congo Red agar include Coomassie blue. It is my understanding that the cellulose will bind the Congo red and the curli fimbriae will bind the Coomassie blue. The use of both dyes will allow for the differentiation of strains based on these characteristics. The resulting phenotypes are rdar (red, dry, and rough; cellulose +; curli +), pdar (pink, dry, and rough; cellulose +; curli -), bdar (brown, dry, and rough; cellulose -; curli +), and saw (smooth and white; cellulose -; curli -). If you have not already read them, I think the attached papers may be helpful for you.
With regard to the incubation conditions, I think testing both temperatures would be beneficial. You may want to consider incubating the plates for longer periods to see how the colony morphologies change over time.

I agree with Emily. Our lab commonly uses agar plates supplemented with only congo red to detect cellulose production on solid medium from Komagataeibacter (Gluconacetobacter) xylinus.
Congo red binds curli, not cellulose. If you want to visualize cellulose, you can use Calcofluor-white. The macro-morphological structures emily is describing (wrinkled/rough) are described well in the attached paper, as well as the differences between the stains. I've been growing Nissle biofilms just fine using these stains according to the attached paper.
