Has anyone here used Cultrex Basement Membrane Extract, PathClear (Cat. No. 3432-005-01) for feeder-free iPSC generation via Sendai virus and for subsequent passaging/maintenance of iPSCs?
I know that R&D Systems offers the Cultrex Stem Cell Qualified RGF BME (Cat. No. 3434-010-02) specifically validated for pluripotent stem cells, and we are planning to obtain it. However, at the moment I only have access to 3432-005-01 in the lab.
My question: Is it worth trying with 3432-005-01 for iPSC reprogramming and early passages, or is it strongly discouraged compared to the stem cell qualified formulation?
Any experiences, tips on coating/dilution, or references to published work using 3432-005-01 in this context would be greatly appreciated.

No, Cultrex Basement Membrane Extract, PathClear (Catalog #s 3432-005-01) is not the recommended product for iPSC generation and maintenance. This is a general-purpose matrix that contains higher levels of growth factors, which can cause stem cells to differentiate and lose their pluripotency. The iPSCs must be maintained in a specific, undifferentiated state to retain their "stemness" and their ability to differentiate into various cell types. The presence of high concentrations of certain growth factors can trigger differentiation signals, causing the iPSCs to lose their desired state.
By contrast, Cultrex Stem Cell Qualified RGF BME (Cat. No. 3434-010-02) which is a reduced growth factor (RGF) formulation minimizes these unknown variables, providing a more consistent and defined culture environment necessary for reliable stem cell research.
So, it is not worth trying with 3432-005-01 for iPSC reprogramming and early passages.
