I am attempting protein expression using the PichiPink system. I've noticed that the cultures of untransformed cells are not always pink, even when the cells used to start those cultures (coming from single colonies on YPD agar plates or glycerol stocks) are pink. Could that be because the YPD medium contains/supports the production of adenine initially? If you proceed with the transformation and plating of these whitish cells, can you eventually tell the difference in color when you plate them out on PAD agar plates?
