My Oil Red O stained mouse fatty liver has more white vacuoles than red stains (Oil Red O). Does anyone have an idea what could be happened ? Some of my slides were really nice. But still I notice the vacuoles significantly. Are the fat is removed when washing ? (protocol in brief, 10um cryosections, fixed in 10% formal calcium, 2 times dip 5 min per each in 100% propylene glycol, Oil red O, 10 minutes agitating, 3 min in 85% propylene glycol, wash in distilled water, counter staining with Harris Hematoxylin, 3 min. wash in tap water and distilled water)
