Hi,
Im trying to do an MTT assay with UT7 cells ( in suspension). I start my asaay treating the cells with different EPO concentraions and another inhibitor. There are 4 96 well plates- 0-72 hours. My stop solution consists HCl- Isopropanol solution.
My question is did anyone had a phanoman in which after adding the stop solution certain well become turbid? it obviously affects the readout.

Using acidified isopropanol as the solubilization solution can provide stable absorbance only up to 20 mins. So, if you are using HCl- Isopropanol solution you need to record the absorbance reading at the earliest.
Another way out would be to use DMSO as the solubilization solution as consistency is maintained even up to 1.5 - 2 hrs after the addition of DMSO. Please refer to the paper attached for more information>
