It is known that Cisplatin (CDDP) (HY-17394, MCE) is a platinum-containing anti-tumor drug and belongs to a polar complex. Why do you recommend me to use water or DMF as a solvent instead of DMSO?
Have colleagues tried the DMSO dissolution system in experiments? If it is feasible to dissolve, what conditions should be paid attention to (such as concentration and temperature)?

Based on the literature (PMID: 24812268), DMSO can inactivate Cisplatin through ligand substitution reaction.
Specifically, the sulfur atom in DMSO will coordinate with the Pt2+ of Cisplatin, replacing its chlorine ligand to form a new complex (such as the single DMSO substitution of m/z 343 and the double substitution of m/z 404). This structural change will significantly reduce the cytotoxicity of Cisplatin.
Experiments show that the IC50 value of Cisplatin in DMSO is more than 40 times higher than that of the saline system, and its ability to induce DNA damage (such as γH2A.X focus formation) and cell apoptosis (such as PARP and Caspase-3 cleavage) is significantly weakened.
