Differences between Calcein (Cat.No.: HY-D0040) and Calcein AM (Cat.No.: HY-D0041):
Calcein AM (Cat.No.: HY-D0041) can penetrate the cell membrane by passive diffusion because it contains an acetylmethoxy (AM) lipid-soluble modification group.
Its molecular mechanism is as follows: the AM group shields the polar carboxyl group of Calcein to form a neutral lipid-soluble molecule, which significantly enhances the permeability of the cell membrane; after entering the cell, it can be specifically hydrolyzed by intracellular esterases, removing the AM group and releasing Calcein with fluorescent activity, thereby achieving specific intracellular fluorescent labeling.
Calcein (Cat.No.: HY-D0040) lacks AM modification, is hydrophilic and negatively charged, and cannot penetrate the cell membrane autonomously, so it is only suitable for extracellular labeling or cell staining after permeabilization.
