Recently, when I was establishing a cell senescence detection system, I needed to select specific marker dyes for senescent cells for different experimental purposes (such as real-time tracking of living cells, high-throughput screening of fixed cells, or staining of tissue sections).
I hope to ask for the experience of colleagues in the field.
The labeling strategy of senescent cells can be based on morphology, function or molecular markers. You can choose the appropriate dye or kit according to the research needs of the subject.
MCE dye products are recommended as follows:
1. β-galactosidase staining kit (Cat.NO.: HY-K1089/HY-15934), which can detect the marker β-galactosidase (a more common marker).
2. P53 (Cat.NO.: HY-P80938), P21 (Cat.NO.: HY-P80774) and P16 (Cat.NO.: HY-P80615) antibodies can be used to detect the upregulation of P53, P21 and P16 (markers of cell cycle regulatory proteins).
3. γH2AX antibody, which detects the upregulation of γH2AX, a marker related to DNA damage.
4. There are some other indicators. You can look at the tweets I posted before, but different aging models will be a little different. It is best to look at the specific cell lines and models and refer to the literature for specific recommendations.
