I am working on the relationship between tissue damage repair and cell pyroptosis. The cell model I used were THP-1(human) and BMDM (mouse). This is the way I use to induce THP-1 cell pyroptosis: PMA treatment (200ng/ml) for 24 hours; change to fresh 1640 (without FBS) for 24 hours; stimulated with LPS (1 ug/ml) 3 hours; then adding Nigericin (10 uM). Previous induction results have been pretty good, WB can detect clear cleavage of caspase-1 and GSDMD, until recently I changed the culture medium and serum form different companies. Suddently, I can not detect the cleavage band anymore. At first, I didn't think it was a problem with the culture medium or the serum. I tried many times until I used the original serum and the cleavage band appeared again. Therefore, I wonder what's wrong with my cells and why changing the culture medium or the serum will affect the induction of cell pyroptosis? Can anyone give me some advices?
