I will appreciate if someone could give me a detailed protocol for fixation adn embedding adipose tissue for transmission electron microscopy, in order to see mitochondria cristae.

In our lab we did a colaboration with a facility that performed the protocol as above described:
Adipose tissue (~1 mm3) was fixed with 2% glutaraldehyde for 2 h,
followed by 1% osmium tetroxide for 2 h and 0.5% uranyl acetate over-
night. After dehydration and embedding in resin, ultra-thin sections
(90 nm) were stained with uranyl acetate and lead citrate and analyzed
in JEOL 1010 transmission electron microscope.
