Transfer for alpha-actin 42kda protein?

HI all,

What is the ideal conditions for protein transfer for a 42kda protein? I use the Bio-Rad mini Blot transfer system with Bjerrum Transfer buffer. I usually run transfers at 0.2A for 90 mins, but I am concerned I will over transfer the protein and lose it. Any suggestions?

Dear Ashley,

The proteins with molecular weight between 30 - 100 kDa are rather easy to transfer without overblotting. I usually use 35V overnight or 1h for 60 min. Ofc in 4°C, good luck!
