i want to ask why is the total phenolic content (TPC) and the total flavonoid content (TFC) of the Ethyl acetat fraction is higher than thos in the Crude Methanolic extract ????
May because of the molecules are more soluble in ethyl acetate which is present in your plant materials, hence it is showing more phenolic content. After ethyl acetate extraction, the left over content might have came to methanol.
Ethyl acetate is intermediate polar solvent as compared to methanol (highly polar solvent). The phenolic and flavonoid compounds present in the plant you are extracting might be of intermediate polarity and showing more solubility in ethyl acetate than in methanol. Also if you are saying that ethyl acetate fraction is obtained from the methanol extract then because of the enrichment of the intermediate polar phenolics and flavonoids you may be getting higher phenolic content with ethyl acetate extract.
Yes the ethyl acetate fraction is obtained from the methanol extract, let's say that the methanolic extract contain 100% TPC, this percentage should be devised between the different fractions according to the polarity of the solvent ? right ? but i am getting like 200% TPC in the ethyl acetat fraction ???
i found a couple of articles with same results but no one gave a good explanation ??
Can you share those articles? It might be the different way of expression for the concentration say e.g. concentration based on the dry weight of methanol extract or concentration in the liquid form or else solvent might be interfering with the analytical method.
