I would like to ask about the way to estimate total nitrogen and I tried with persulphate method where in I use 30 mins autoclave condition for digestion and after the conversion of all N in the sample to nitrate I did the nitrate estimation through chromotrophic estimation method. However I did get result for urea standard but for ammonia standrad no results were observed. So can anyone suggested with there experience what other way to find total nitrogen or any alteration in the persulphate method I used.

Thanks Dr. Monyer for the link, but my problem is i have a liquid matrix for which total nitrogen have to be determined. Secondly any colorimetric method that can be used for total Nitrogen estimation.
The one method I mentioned it works by digesting any form of N into nitrate using persulphate which then reduced to nitrite through cadmium column and total nitrogen value will be estimated... however i tries doing that method i got results for urea standard that it gets digested and converted into nitrate but ammonia didnt get converted into nitrate.... so this is my manor problem....
You can find a very detailled information about nitrogen forms in soils and their determination in Methods of soil analysis. The pesulfate methods enable to determine total nitrogen to some extent but it comparison to the other commonly used method has not been investigated in detail. In persulfate methods Devarda alloy is used to reduce nitrate to ammonium (Raveh and Avnimelech, 1979; Bremner, 1996. Nitrogen Total. in Methods of soil analysis, pp 1085-1022. In a similar way you can use distillation methods, if you have disttilation aparatus, as in the case of regular Kjeldahl method after reducing the nitrate-N to NH4. Then the N content of the extract can be colorimetrically determined. There is another persulphate method using potasium persulfate and NaOH as digestive reagents (Huang and Yang, 1987. New method for determining total nitrogen in soils and fertilizer, but this is a chinese paper.
