In the total coliform bacteria test, a fine porosity filter is used to retain bacteria. The filter is placed in a petri (culture) dish on a pad with growth enrichment media (mEndo) and is incubated for 24 hrs at 35 degrees C. subsequently the plate i counted for most coliform forming units. A confirmation test using more specific media is usually followed.
1) Can i replace mEndo broth with LB/R2A? would there be any consequences in doing so? (since these broths are non selective)
2) For the confirmation test which broth is recommended?
For research, stick with standard method for analysis of water and wastewater (book). Millipore Corp is one of the companies selling supplies. When dealing with unknown contamination level, you might have to dilute several times to avoid overcrowding and get countable colonies, especially when dealing with stormwater or contaminated Wastewater.

Please check out the link I'm sending you. There is a lot of information (specifically for US regulated pharma) maybe you can find something that apply to your work. I have used mEndo for coliform test in water samples (recommended), and R2A to evaluate other type of bacterial recovery for water samples, both MF test. Also, I have used MacConkey broth (42-44C) and agar (30-35C) for absence of E. coli but, for specific Raw Material samples, not water (incubation time varies from 18h-72h). If you are doing research you can try different conditions and assess results to come out with the best outcome for your purpose. Is there any particular reason you want to change media type?

Hi, you can use LB media in the case of the MPN method for total coliform. for the conformation, you can use Mackonkey Broth.
