Hi . I am a student researcher and I am writing this to request your help in obtaining Top 10 cells and expression plasmid for my research experiment. Unfortunately, my current budget does not allow me for the purchase of these reagents and I am hoping that someone may be able to help me

I think that Kulsoom Aftab is asking for the Top10 (strain name) bacteria for transform and some expression vectors (plasmids).
Have you considered other transformation methods (LiAc, calcium phosphate or electroporation) or making your own chemically competent bacteria? You can often find pUC18 and pUC19 plasmids in lab freezers as they often come with kits as a control.
What expression plasmid are you planning to use? You may not need to use Top 10 cells but might be able to use many other E. coli hosts instead. What options do you have available and maybe someone here can tell you if they would work. Top 10 is not really that unique a strain.
