I know for following the potency of our mature dendritic cells differentiated from monocytes, we should check the proliferation of the outologe T cells, but is it necessary to test the potency of mature dendritic cells to induce the proliferation of someone else T cells?
I have another question. Is there any need to add IL-2 to the medium of DC and T cell co-culture. If yes, IL-2 doesn`t effect on the exact and real response, doesn`t it cover the result in which the function of potent DC of interest is going to be considered?.

One of the hallmarks of matured DCs is to induce the proliferation of T cells. This can be done in an allogenic fashion (DCs and T cells from different donors), where you make use of MHC mismatch. An autologous system (DCs and T cells from the same donor) will require antigen-specific stimulation and antigen specific T cells.
As for IL-2, you don't need to add it, especially if you are working with CD4+ T cells, which will start producing IL-2 themselves shortly after stimulation. Using IL-2, especially at high concentrations, may interfere in T cell polarization and favor the priming of Treg cells.

Again, I want to add this note that according to what I tested in lab, the dosage of more than 120 IU/mL of IL2 will definitely expand the number of T cells, but in many articles i have read that even 300 IU/mLof IL2 was added to the co-culture of DC and T. I myself was hesitated to apply this amount whenever we are performing the tests which we are not sure about the intensity of immune response. But as I intend to continue the exposure of DCs to T cells for 14 days, I think it is better to add the low amount of IL2 for the maintanance of T cells. In the meantime, by studing plenty of relative and associated articles in this concept, I reach this amount. I will be more than glad to know and be informed of your experiences.
