The idea sounds OK if you are sure that body position does not change between those contractions with the same weight, Other issues you would have to deal with are if fatigue or injury occurs during the contractions. In those situations, sEMG amplitude would be increased even if you are lifting the same load.
Yes, it seems the right thing to do (and the above answer). Also since Biceps brachii operates through a wide range of elbow angles, (different lengths), it is worth keeping that in mind too!
