Hi, I am trying to polarize human monocyte derived macrophages (M0 cells cultured with medium only for 5 days) by IFNg and IL-4. The methods from papers are always for 48h polarization. I am wondering if I can prolong a longer time, culturing cells with IFNg or IL-4 for e.g. 5 days? Thanks so much for your help!

It is known that 48 hours is quite sufficient time for M0 cells (M2 markers need more time, even 48h) to prime the signal from IFN-γ/IL-4 and polarize in the direction M1/M2. As you know for many reasons, it is important to shorten the period of manipulation of cultured cells. After this period, these polarized cells can be used for different experiments. In my opinion, as far as the comparison of produced cytokines is concerned, it is important to compare them at same time period.
