I have been having some issues lately with the PureLink plasmid giga prep kits from Thermo Fisher.

They had been working well for quite a while and then all of a sudden i started having issues when trying to pass the lysate through the column using suction. It seems the pressure builds up inside the bottle and then bubbles start going through the suction port.

There are lots of bubbles in the column, and i have been equilibriating the column first. If you bang the column enough to get all the bubbles out, it eventually all filters through but in the end, there is no plasmid at the end of the protocol.

I noticed a bit of rust at the suction tap (copper filter or something?) i changed that and everything was going perfectly for a week. Today i did two preps, the first one worked well and then the second one went back to the bubble issues.

Has anyone else had this same issue and fixed it?
