The time point of UVB-induced inflammation?

I'm studing the UVB-induced inflammation in HaCaT cells, however I'm not sure how to choose the correct time point. Should I pretreat cells with LPS ? After irradiation, how many hours should I wait for protein extraction to assay the IL level?
