The pSa ori needs a functional repA gene to operate and usually is for low copy replication in Agrobacterium. I assume your plasmid is a Ti plasmid? the other ori is for cloning and maintenance in E. coli.
No, it is not impossible. But what Robert Adolf Brinzer is saying is that the plasmid you refer to has two origins but only one works in Agro and only one works in E. coli.
However it is possible to have multiple origins in a plasmid. If you think about it, any plasmid dimer will have 2 origins.
Thanks, I got it. It is not Ti plasmid, which should be a plasmid dimer. The different origins functional on different host strains. But I know the pSa ori could also work in E coil. Maybe in E.coil, colE1 ori works preferentially.
