I've been using 1µg/µl LPS and injected 2µl into each SN stereotaxically, according to accessible articles protocol. unfortunately rats died immediately after two-side injection of LPS. However some of one-side injected rats were failed too.
If anyone could figure out the reason, I would be appreciated to inform me.

I would like to ask you two question to have more information and, then, try to figure out what is happening.
- Which rat strain are you using?
- Could you know the total Endotoxin Units that you are injecting?

LPS is notorious, there are many types and sources can wildly vary. It most likely caused a type I allergic reaction. You should have done a post-mortem examination of the rat to find out the cause of death.
It is not a good model for Parkinson's. There are many other models of Parkinson's available. Please read a few good reviews such as:
Evaluation of Models of Parkinson's Disease; Front Neurosci. 2015; 9: 503.
Personally, I like the Rotenone model because it is a naturally occurring compound and the effects are quite clear. It is also quite simple and do not require complicated surgery. Just make sure you get clean results under your laboratory conditions with your specific rats.

I chose SD rat and injected 2µl lps ,it seems even 0.5µl lps could be fatal !!
I saw intracranial bleeding in some cases.
They couldn't recover after surgery and heart rate was high . They were breathing irregularly.
LPS -induced Parkinson seems to be more immunological and I want to evaluate the role of inflammation and NLRP3 activation in PD model.
