For example, a natural compound named ruin have IC50 value reported at 100microMolar concentration in HCT-116 cell line in one research paper, while the value differs to as low as 40microMolar in another research paper and value increases upto 200microMolar in yet another research paper. What is the technical reason behind such a fluctuation in IC50 value evaluated through cytotoxicity assay?
The purity of the compound could be one factor, if it varies. Another factor could be the precise growth conditions, such as the FBS percentage in the medium, especially if the compound binds to serum proteins. A 3rd factor could be variation in the calculation of the IC50.
In fact that level of variation between reports is not too bad, at just over 2-fold in either direction from a value of 90 µM, 5-fold range from top to bottom. Experimental variation in the measurement and the aforementioned factors could easily account for such a range.
This is very common based on my experience. The wide range of IC50 values for a specific natural compound in different studies and cell lines can be attributed to several factors:
Compound Purity: The purity of the natural compound used in experiments can vary between studies. Impurities or degradation products in the compound can affect its activity and lead to variations in IC50 values.
Experimental Conditions: Variations in experimental conditions, such as culture media, incubation time, temperature, and drug exposure duration, can influence the IC50 value. For instance, natural compounds often have complex interactions with other molecules present in the cell culture or in vivo environment. The type of media used in each of those studies might have led to such variations if the formulation had differences. Such interactions can influence the compound's activity and result in varying IC50 values.
Methodology: Different laboratories may use slightly different methodologies and assays to measure IC50 values. Variations in assay techniques, such as the choice of assay reagents, detection methods, and data analysis, can lead to discrepancies in IC50 values.
Publication Bias: Studies with statistically significant or interesting results are more likely to be published, leading to potential publication bias. Studies reporting extreme IC50 values may be more visible in the literature, contributing to the perception of a wide range of values.
There can be lots of reasons e.g.,
1) The concentration of the cells. Many toxic agents show a cell concentration dependency, this can occur if the cells metabolize the compound to a more (or less) cytotoxic compound. This is observed with hydrogen peroxide. Thesis Nitrofurazone and Hydrogen Peroxide Metabolism in Trypanosoma brucei.
The higher the concentration of cells, the more rapidly the peroxide is removed, and the less overall cytotoxic stress experienced by the cells. Conversely if a compound is metabolized to a permeable cytotoxic species, the more cells the more toxicity.
Another cause can be the growth phase (logarithmic, or plateau phase) the cells are in (or just their overall growth rate) for example many anticancer drugs show higher toxicity to rapidly dividing cells.
Other factors could include the media's pH or the levels of nutrients remaining in the media etc.
