I plan to use a UV lamp to create DNA breaks in the cells. The UV lamp in our lab is fixed in the cell culture hood (Ushio germicidal bulb, UVC, wavelength 253.7 nm, 15 watt, 55 volt) and I need to introduce 5J/m2 to my cells.
As Watt= Joule/sec, it seems that to have 5J/m2 from a 15W bulb I just need to consider the time I expose my cells in UV(sec) and the m2. I'm not sure the m2 comes from the (square) distance from the UV source to my cells, or from the area of the surface that receive the UV light?
Is that way of calculation appropriate? (I read around and people mentioned many other factors (even consider the length of the lamp?) and different types of other complex formula that I don't quite understand)
