I am planning to test the efficiency of designed gRNA by performing a T7 assay, and I was wondering whether someone would have a protocol for Lipofectamine™ 2000 transfection of NIH-3T3 cells? I would not need high transfection efficiency, but rather to check for the formation of structural deformities in DNA heteroduplexes.
In addition, I was wondering whether someone has a suggestion of some other cell line that I could potentially use? My gRNAs are designed against mural targets, so human cell lines would not be an option. Thanks!
