Hi all, I'm starting to work with AAV. The protocol that I follow purify AAV by iodixanol gradient ultracentrifuge. The rotor that the protocol recommended is VTi 65.2 Vertical-Tube Rotor.

However, I only have access to a swing bucket rotor (SW 41 Ti Swinging-Bucket Rotor) to accommodate required speed. Can I use the swing-bucket rotor for my procedure without any changes to speed? Will swing-bucket rotors maintain density gradient after the centrifugation process?

Additionally, I have to increase the scale for my SW rotor (from ~5 mL to ~10 mL). Can I simply double each volume fraction that I pour into the tube without changing the speed? I wonder whether the liquid column height have an influence?
