I am running a Western Blot Analysis of a FAT1 gene knockout that I established in an epithelial cell line. I have performed the Western Blot once, using a gradient gel. However, my protein of interest is very large (about 500-600 Kd). At the top of my gel, it looked like I had a band for my positive control and none for my KO sample. However, my ladder only goes to 250 Kd, so I can't confirm if that is my protein of interest. I have checked several distributor websites looking for a bigger protein ladder, and unfortunately, I can only find 1 Kb DNA ladders. Does anyone know where I could find a bigger protein ladder?? Thank you.
First, are you trying to do PAGE/Western blot or nucleotide analysis? The first requires Protein Ladders with kDa scale, the latter requires polynucleotide marker with bp scale. For Western blot try to look for Protein Ladders. eg. HiMark from Thermo goes up to 460 kDa.
