I’m working with an mRuby-tagged beta2 AR plasmid, which previously showed consistent membrane localization. However, I’m now observing increased ER retention. I’ve attached Picture 1 as a reference for the original membrane expression, and Picture 2 shows the current expression pattern. Nothing in the protocol has changed.
Any insights on what might be causing this change or suggestions for troubleshooting would be much appreciated.

The cells in image 2 look like they are rounding up. Once your cells start to peel up, the localization is going to go awry. There may be other things going on, but this is what jumped out at me. It could be due to passage number, confluency, etc.
