I am trying to do helical reconstruction for my fibrils. I am at 3D classification steps where I can get the resolution (given by relion) at 3.2 A. The actual resolution is worse I think. Anyway, I am advised to try autorefine feature of relion but the lowest I can get to is 4.6 A. I have tried the following:
- Release the rise
- try to Fit in Map the two halves using Chimera and rerun the autorefine
- Try to get the z-percentage to 0.1
- manipulate the asu unit
But nothing goes pass 4.6A.
I wonder if there is a stepwise procedure/instruction that I can follow? Any suggestion is very appreciated.

I did this back in 2014 and 15 when Relion was still in its infancy and did not have a helical pipeline. A lot of errors came from a seeming bias to make a spherically masked particle and so refining our angular search parameters in the source code was our olnly option; however I believe this has been refined in the code. All I could recommend is Article Helical reconstruction in RELION
and emailing Sjors Scheres directly. Also recommend emailing Benjamin Himes at Janelia Research Institute in the Grigorieff Lab directly with your question.It may also be that 4.6 is close to your resolution limit.
