A lot of papers cultered number of 1 million stem cells per one injection in vivo, can I culture lower number of stem cells? I got a yield of 300 000 cells only, how will the low number affect the results? Or if can I depend on its natural feature of self renewal in vivo?
It may just be a matter of time to allow the cells to grow. Growth rates depend on culture conditions and cell type. If you examine your culture under a microscope you may be able to detect dividing cells by looking at their nuclei. If not a careful examination of the culture plate at different times may reveal filled-in areas that were previously empty of cells. Confluence is reached when the entire plate is covered by cells. Contact inhibition may stop the cells from dividing further. You probably do not want to let a confluent culture sit too long before you harvest the cells to "split" the plate unless doing so is part of your protocol.

Please describe your work with more detail. What type of stem cell are you working with? What do you want to do with those cells exactly?
You must know that when the cells start to differentiate, the rate of their amplification decrease. Hence, in many clinical trials or in vivo examinations, expansion of the cells is considered in vitro (before in vivo step) to obtain adequate population of the cells before in vivo phase.
The number of MSCs to be infused depends on many variables (i.e, model of disease, source of cells, route of administration, number of doses). In my experience, I have shown good efficacy even with
