Hi,
I am having trouble marking the calcium deposit of osteoblasts from the MC3T3-E1 Sub 14 lineage. After staining with alizarin red, it is not possible to observe the presence of mineral deposits. Cells are ALP positive in situ stained with Fast Red.
My question is regarding the pH of alizarin red. Should it be 4.1~4.3 or closer to alkaline? And what is the best way to prepare the solution? Just dilute the powder in deinized water (room temperature)?
