Considering the difficulty in ensuring consistent and homogenous seeding of cells in multi-well dishes and the endless number of threads/advice/voodoo people resort to in order to avoid the cursed clustering of cells in the middle. Why doesn't anyone manufacture multi-well dishes with square wells? Is there an inherent reason? A practical one?
From my experience, for example, large square dishes and rectangular flasks give much more reproducible results than traditional round dishes.
also, leaving the plates in the hood for about 30 minutes before placing in the incubator reduces this edge effect. PMID 14567784. We also use the square wells for other experiments (miilipore also makes culture slides suitable for imaging). We have had some issues with different manufacturers with leakage and wicking between wells, however
David, thanks for the correct search terms :-) I have requested samples of the Nunc plates, let's see how my cells like them.
Andrew, thanks for the reference. I already try and leave the plates on a solid bench away from users (the hood is almost constantly in use), but some of our cell lines attach very slowly, so even after 4 hours any movement to the plate can cause swirling and clustering. I will have a look at Millipore's products too.
