Someone help me to understand TCS haplotype network please?

I need to generate haplotype networks for 28 S sequences of insects. I already used POPART but i need to learn how to insert and write in the nexus file groups such as host, location ect.. I need also to learn how to use TCS and statistical tests like AMOVA, Tajima's D ....
