I am wondering, If there is some possibility to solve NMR spectra of multiple DNA conformers (such a mixture of parallel, hybrid and antiparallel DNA G-quadruplexes) in a single solution (sample). Does it require some modification of common NMR procedure or is it still insoluble problem? Because it is not feasible to separate DNA conformers of the same length before NMR.

By applying NOESY yada NOE technique you can have information about spatial orientations.
If one of the is the major conformer making up ca 80 % of the sample you can potentially do it. If the conformers are of almost equal concentration I would seriously doubt that the solution of all the structures would be possible. Of course it also depends on strand length. For a VERY short piece it might work.

my DNA oligos are about 30 nucleotides long. Unfortunately, the concentration of conformers are almost equal, so probably I will have to optimize protocol of quadruplex induction to obtain only one main conformer for further NMR investigation.

you talking at least 3 conformations. You either find point mutations that will prevent one of the conformations to form or to be really minor or you find a salt mixture not favorable to oe of the conofrmers or you find a temperature that melts one conformer before the other. But the three at same time its a no, you cannot.
