I am trying to stain CD68 and CD11b on tumor tissue (mouse).

I have collected mouse tumor tissue, fixed it in 4% Paraformaldehyde, dipped it in 30% sucrose and cryo in OCT -80oC.

The tissues were sliced and fixed with cold acetone (or methanol) before staining. The problem is ONLY CD68 can be detected, not CD11b.

So I changed the protocol with antigen retrieval (heat 90oC in Sodium Citrate Buffer pH6.0 or Tris-HCl pH9.0 buffer) but the result was ONLY CD11b can be detected, not CD68.

Do you have any experience with this problem? I just want to detect both of them on one slice.
