As you describe in your question is quite confusing, but in my opinion you should prepare 10X HBS with 1X sodium bicarbonate. May be you need just 10X HBS but not 10X sodium bicarbonat.

Please, clearly mention the sentence what you are trying to follow for preparation.

Thanks for your response.

Actually I am trying to develop 3D collagen gel embedded with fibroblast. I came across few protocols in which they add 10X HBS (with phenol red) to collagen solution (acts as pH indicator) and after which few drops of NaOH is added to neutralize the pH.

So I have HBS powder in my lab and wanted to prepare 10X solution out of it. My question was should I supplement it with 10X of sodium bicarb and phenol red or both of these to be added acc to 1X??

Okay now i understand your question clearly.

You have to prepare 10X HBS and other contents (Sodium bicarbonate and phenol red) should be acc to 1X.

And you are right excess (if 10X) phenol red is toxic to cells, go for just 1X.
