The paper below knocks out both allele of B2M gene using adenovirus.

They have targeted B2M allele by transducting adenovirus sequentially using two different antibiotic selection gene. Why do they have to target it twice? idelaly, single transduction should work since homologous region are same in both allele. If I have to do homologous recombination for a particular gene, do I have to generate two virus with different selection gene? Why should single transduction work? I need an urgent help of scientific community.
