Hello all!

I am currently working with the single-molecule force spectroscopy using AFM (Multimode scanning probe microscope with a Nanoscope V controller (Bruker Inc.)) under constant mode to study the protein unfolding. I have my AFM tip modified with streptavidin to pick the biotinylated proteins. I have a few questions regarding the force curves obtained.

1. The force curves are not stiff instead it has a wavy nature (look image below). I have a doubt whether I am having the correct force curve or not.

2. There is an increase in the contour length with the NHS-PEG maleimide treated surface (Protein length=278 AA). I have 2 fusion proteins anterior and posterior to the protein of interest (POI).

3. Missing the PEG stretching curve in the control samples.

Could someone clarify me with these queries?
